Supplement to: Dunckley T et al. Whole-Genome Analysis of Sporadic Amyotrophic Lateral Sclerosis. N Engl J Med 2007;357(8):775-88. DOI: 10.1056/NEJMoa070174.

This appendix has been provided by the authors to give readers additional information about their work. Data is also available from the authors at www.tgen.org/neurogenomics/data.

Available clinical data for all samples genotyped in both the Discovery and Replication groups. The header row indicates the specific information detailed in each column.

Complete rank ordered SNP analysis from the Affymetrix 500K Mapping Arrays. SNPs were rank ordered as described and referenced in the Methods. The top 192 SNPs were chosen for genotyping in the independent validation population.

Complete rank ordered SNP analysis from the Illumina Infinium II HumanHap300 bead chip arrays. SNPs were rank ordered as described and referenced in the Methods. The top 192 SNPs were chosen for genotyping in the independent validation population.

Complete odds ratio calculations for heterozygotes and homozygotes for the SNPs presented in Table 1. Calculations were made using the DeFinetti program (http://ihg.gsf.de/cgi-bin/hw/hwa1.pl) and methods were adapted from Sasieni9. The first two tabs of the spreadsheet contain calculations for all replication series 1 Caucasian ALS patients versus all Caucasian controls and mixed ethnicity ALS (including Caucasian) versus the Caucasian controls, respectively. A legend detailing the abbreviations used is in the fifth tab of the spreadsheet. Columns, from left to right, show the SNP tested, Hardy-Weinberg equilibrium tests for cases and controls, the odds ratio calculated using the allele frequency difference between cases and controls, the odds ratio for heterozygotes, the odds ratio for homozygotes, the allele positivity odds ratio, and the common odds ratio calculated using Armitage’s trend test, along with statistical tests for significance of each. Only the Caucasian ORs and P values are referenced in the text.
