To the Editor: Bisphosphonates are widely used to inhibit osteoclasticbone resorption. As analogues of pyrophosphate they share severalstructural homologies with recently identified non-peptide /T-cell ligands, such as isopentenyl pyrophosphate, that activateantigenic receptors of / T cells.1
We recently found that aminobisphosphonates (pamidronate, ibandronate,and alendronate) can stimulate the major subgroup of / T cellsin peripheral blood (V9V2+ T cells), whereas non-aminobisphosphonates(clodronate and etidronate) are inactive. In our assays, pamidronate(concentration yielding half-maximal activity, 5 to 10 µM)induced the activation and selective expansion of / T cellsin cultures of peripheral-blood mononuclear . . .
